Background:Crestor is a widely used drug for the treatment of benign prostatic hyperplasia (BPH) in adults and adolescents over 40 years of age, but its long-term use remains controversial. The aim of this study was to compare the drug’s efficacy and tolerability in treatment of BPH.
Methods:This was a randomized, double-blind, parallel-group, parallel-group, multicentre study conducted at the Faculty of Pharmacy, University of Cape Town, South Africa.
Results:In the overall population, 10-mg dose of rosuvastatin was associated with significant reduction in the duration of follow-up, duration of treatment, and reduction in the total number of active symptoms, and reduction in the frequency of new symptoms in the group treated by rosuvastatin.
Conclusion:Rosuvastatin, with a dose of 5 mg, is effective and well tolerated in patients with BPH.
Rosuvastatin is available in the USA:Table 1
Rosuvastatin is used in the treatment of patients with BPH. Rosuvastatin is also used in the management of menopausal symptoms, and for treatment of BPH in women. There are no published reports on the efficacy and safety of rosuvastatin in women with BPH. Rosuvastatin is a once-daily oral drug which should be used regularly in women with BPH and who are already taking oral medications.
Table 2
The dose of rosuvastatin is 20 mg daily. The drug is indicated in the treatment of benign prostatic hyperplasia (BPH) and in the management of male-pattern dysfunction. In addition to the initial dosage, rosuvastatin should be administered daily in conjunction with other medications for the treatment of the above conditions.
Key words:Benign prostatic hyperplasia, rosuvastatin
Patients and Methods:This is a single-center, double-blind, randomized, parallel-group study comparing the efficacy and safety of rosuvastatin in patients with BPH. A total of 80 patients with BPH were randomized to receive 20 mg of rosuvastatin once daily or placebo. Patients with active symptoms at baseline were included in the study. A total of 8 patients with BPH and in whom active symptoms were present were included in the study. Rosuvastatin doses were based on the maximum recommended daily dose. Patients were divided into two groups (n = 80): rosuvastatin 20 mg or placebo (n = 80); rosuvastatin 20 mg plus placebo (n = 80). Patients were monitored regularly for signs of BPH, symptoms of BPH, and the occurrence of any adverse events (AEs) during the treatment. Safety was assessed using the intent-to-treat analysis.
The mean age of the patients was 45.6 years, and the mean body mass index (BMI) was 28.9 kg/m2 (range 18.5 - 44.1). The majority of patients had moderate to severe hypertension (defined as a BMI < 40 kg/m2 or < 25 kg/m2), with a low to very low risk of BPH. The most common adverse events (AEs) were headache, dizziness, dyspepsia, nausea, abdominal pain, and insomnia.
In the rosuvastatin group, the mean dose of rosuvastatin was 40 mg once daily, whereas in the placebo group, the mean daily dose was 5 mg. The frequency of AEs was significantly higher in the rosuvastatin group (17.3 vs. 11.2 per 100 person years) (P < 0.05). The most common side effects of rosuvastatin were headache, dizziness, nausea, abdominal pain, and insomnia. Conclusions:Rosuvastatin is a well-tolerated drug for the treatment of BPH, but its long-term use remains controversial. Therefore, there is need for more studies in patients with BPH and in the long-term treatment of BPH.
